if you just focused on the idea and didn't so much write the exact plot it would work LOADS better. Yeah you're right. It would be fun to borrow small elements of canon, but it totally needs its own overarching plot.
I can see Dean's attachment to Sam being a precursor of his attachment to Cas. Angels are supposed to be aloof, and they're supposed to love all their brothers and sisters the same, but Sam and Dean have a special affinity for each other. Dean maybe feels a little guilty about it, but Sam embraces it more, and maybe his affection for Dean is partly what inspires Sam to fall in the first place, because loving Dean opens Sam's eyes to this whole new world of ~feelings~ or whatever. Maybe he sees that Dean is skittish about ~feelings~, and though it hurts Sam to have to leave him behind, he won't force Dean into having to choose his loyalties. He knows how loyal Dean is. So Sam rips out his grace, and Dean is totally upset by this. Sometimes he wonders if he would've or should've fallen with Sam, but he always shoves those kinds of thoughts out of his mind. Such thoughts are unbecoming for an angel. But yeah, he still keeps an eye out for Sam on earth. (Maybe looking for Sam is part of his secret agenda of being on earth. He's there for Cas, but also secretly for Sam.)
So then like in canon, maybe Sam has to go find his grace. This can be plot-relevant somehow. What will Sam then do with his grace? Also lol it's kind of funny how even in this universe, angels think Sam Winchester must die. So once Sam angels up and saves the day, he has to go into hiding, but before he does that, he and Dean are going to have a ~moment~. It's gonna be bittersweet, because they just found each other again, but just as quickly they are forced to part.
Also, in this 'verse, I ship Sam/Anna. Also Sam would be hilarious randomly popping into the backseat of their car and freaking them out.
As for how Cas and Anna become hunters, if not their mother burning on the ceiling, then it can just parallel the angel creation story: maybe Cas and Anna are just born into this function. They've been raised in this life since the beginning, it's what they were made for. As for what excuse I'd have for bringing Cas into the croatverse future, hmm, still have to think about that one.
